This region includes a DNase I hypersensitive site that spans an enhancer for the downstream MPO (myeloperoxidase) gene. This sequence is bound by C/EBP, the Myb myeloblastosis viral oncogene, and runt-related transcription factors. A subregion was also validated as an enhancer by the ChIP-STARR-seq massively parallel reporter assay in naive human embryonic stem cells, where it associates with the NANOG transcription factor and is marked by the H3K27ac histone modification.
